Elimination of interfering activity in serum samples in the Chinese hamster ovary pertussis serology assay

An interfering substance in various frozen serum samples was observed to inhibit the adhesion of Chinese hamster ovary (CHO) cells to the microplate surface in the CHO pertussis neutralisation test (NT), resulting in wells lacking cells or wells with dead cells after two days of incubation. The interfering activity of the serum could be eliminated 1) by transferring cells to other wells after initial incubation, 2) by adding foetal calf serum (FCS) to the sample dilution buffer, 3) by precoating microplates with FCS, or 4) by preincubation of samples at 4°C for five days. Preincubation at 4°C for five days reduced the interfering activity in only some of the samples. Addition of serum to the sample dilution buffer or precoating with serum did not influence the antibody titres in the serum samples. The described method may be used for routine applications.
